Topical Cetirizine in Androgenetic Alopecia: A Preliminary Study [PMID: 41625414 · 2025]
Dhurat RS, Sharma R, Shetty C, Mehta N, Goren A, Lotti T · Skin Appendage Disord (2025)

Prostaglandin D2 (PGD2) is elevated in the bald scalp of men with androgenetic alopecia (AGA) and inhibits hair growth. Cetirizine, an H1-antihistamine, reduces PGD2 production and has anti-inflammatory effects, making it a potential therapeutic candidate. Ten men aged 18-60 years with Norwood-Hamilton grade III vertex to V AGA applied 1 mL of 1% cetirizine in propylene glycol/butylene glycol daily for 16 weeks. Exclusion criteria included recent use of minoxidil, finasteride, or other antiandrogenic therapies. Hair counts in a 1 cm 2 tattoo-marked vertex area and blinded photographic assessments were evaluated at baseline and week 16. Nine participants completed the study. Mean hair count increased from 139.6 (SE 13.9) to 163.4 (SE 15.5), a statistically significant change ( p = 0.009). Blinded global photographs showed a +2 improvement in 1 patient, +1 in two, and no change in six. Trichoscopic counts improved in all patients. No adverse events occurred. Despite limitations, this report provides additional real-world data supporting cetirizine as a potential therapy in AGA. Larger randomized controlled studies with longer follow-up are needed to establish efficacy, durability, and mechanism of action.

Prakoeswa CRS, Damayanti, Anggraeni S, Umborowati MA, Sari M, Hendaria MP, Thahir TF · Dermatol Res Pract (2024)

Atopic dermatitis (AD) is a chronic, inflammatory skin condition characterized by eczema lesions and dry, itchy skin. Recent guidelines for the management of AD emphasize the importance of using moisturizers in the management of AD. This study is a double-blind clinical trial to determine the effectiveness of moisturizers containing anti-inflammatory ingredients compared with moisturizers without anti-inflammatory ingredients for skin hydration in mild to moderate adult AD patients for 14 days at the Dermatology and Venereology Outpatient Clinic at Dr. Soetomo General Academic Hospital. There was a significant difference ( p < 0.05) at the baseline and day 14 skin hydration values in the experiment group with anti-inflammatory ingredients (35.97 ± 6.04-66.06 ± 15.84) and the control group without anti-inflammatory ingredients (40.74 ± 10.94-56.12 ± 8.34). After comparison, there was a significant difference ( p < 0.05) in the skin hydration value between the experiment group and the control group on the 14th day. There was also a significant difference in the improvement of skin hydration outcomes between both groups ( p < 0.05). The severity of the disease using Scoring Atopic Dermatitis (SCORAD) showed a significant difference ( p < 0.05) between the experiment group and the control group after 2 weeks of intervention. The addition of anti-inflammatory ingredients in the moisturizer, namely, shea butter, bacterial lysate, allantoin, bisabolol, Phragmites kharka extract, Poria cocos , and Mirabilis jalapa in a moisturizer containing occlusive (dimethicone), humectants (glycerin, saccharide, butylene glycol, and hyaluronic acid), and emollient (shea butter and squalane) was shown to be significantly better in improving skin hydration in patients with mild to moderate AD.

Liu Y, Zhang C, Zeng AP · Biotechnol Adv (2024)

Diols are important platform chemicals with a wide range of applications in the fields of chemical and pharmaceutical industries, food, feed and cosmetics. In particular, 1,3-propanediol (PDO), 1,4-butanediol (1,4-BDO) and 1,3-butanediol (1,3-BDO) are appealing monomers for producing industrially important polymers and plastics. Therefore, the commercialization of bio-based diols is highly important for supporting the growth of biomanufacturing for the fiber industry. This review focuses primarily on the microbial production of PDO, 1,4-BDO and 1,3-BDO with respect to different microbial strains and biological routes. In addition, metabolic platforms which are designed to produce various diols using generic bioconversion strategies are reviewed for the first time. Finally, we also summarize and discuss recent developments in the downstream processing of PDO according to their advantages and drawbacks, which is taken as an example to present the prospects and challenges for industrial separation and purification of diols from microbial fermentation broth.

Microwave-Assisted Extraction of Phenolic Compounds and Antioxidants for Cosmetic Applications Using Polyol-Based Technology [PMID: 39248485 · 2024]
Myat Win S, Saelee M, Myo H, Khat-Udomkiri N · J Vis Exp (2024)

The utilization of polyols as green solvents for extracting bioactive compounds from plant materials has gained attention due to their safety and inert behavior with plant bioactive chemicals. This study explores the sustainable extraction of phenolic compounds and natural antioxidants from coffee silverskin using the microwave-assisted extraction (MAE) method with polyol-based solvents: glycerin, propylene glycol (PG), butylene glycol (BG), methylpropanediol (MPD), isopentyldiol (IPD), pentylene glycol, 1,2-hexanediol, and hexylene glycol (HG). A comparative analysis was conducted on conventional and non-conventional solvent extractions, focusing on their impact on the bioactive compounds of MAE, encompassing parameters such as total phenolic content (TPC), total flavonoid content (TFC), and antioxidant activities like the 1,1-diphenyl-2-picrylhydrazyl radical scavenging assay (DPPH), the 2,2'-azino-bis(-3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging assay (ABTS), and the ferric reducing antioxidant power assay (FRAP). The highest values were observed for TPC with aqueous-1,2-hexanediol extraction (52.0 ± 3.0 mg GAE/g sample), TFC with aqueous-1,2-hexanediol extraction (20.0 ± 1.7 mg QE/g sample), DPPH with aqueous-HG extraction (13.6 ± 0.3 mg TE/g sample), ABTS with aqueous-pentylene glycol extraction (8.2 ± 0.1 mg TE/g sample), and FRAP with aqueous-HG extraction (21.1 ± 1.3 mg Fe (II) E/g sample). This research aims to advance eco-friendly extraction technology through natural plant components, promoting sustainability by minimizing hazardous chemical use while reducing time and energy consumption, with potential applications in cosmetics.

Deciphering trends in replacing preservatives in cosmetics intended for infants and sensitive population [PMID: 39153997 · 2024]
Chen T, Chang H · Sci Rep (2024)

The present study aims to investigate the current trends in replacing conventional preservatives with multifunctional ingredients with antimicrobial properties for preservation of cosmetics for infants or sensitive population, to decrease their potential for contact dermatitis. We first reviewed the labels of cosmetics purchased from the Chinese market for conventional preservatives and multifunctional ingredients with antimicrobial properties, of which the actual contents were further quantified by chromatographic methods. We identified 7 traditional preservatives (phenoxyethanol, benzoic acid (salts), methylparaben, benzyl alcohol, sorbic acid (salts), propylparaben, and methylisothiazolinone), and 11 alternative ingredients with antimicrobial activities (ethylhexylglycerin, butylene glycol, caprylyl glycol, propylene glycol, 1,2-hexanediol, p-anisic acid, hydroxyacetophenone, pentylene glycol, decylene glycol, caprylhydroxamic acid, and aminomethyl propanol) in descending order of prevalence. The contents of all identified preservatives and ingredients were either below regulatory limits or in the range that is generally regarded to be safe. Further challenge with microorganisms indicated irrespective of the composition of preservation systems, product preservation could be compromised under test conditions. We conclude that multifunctional ingredients with antimicrobial properties in cosmetics have the potential to completely replace or significantly reduce the use of traditional preservatives while retaining comparative preservative efficacy. Future attentions may need to be shifted to the safety of those multifunctional ingredients with antimicrobial properties.

Allergic Contact Dermatitis to Topical Preparations Containing Minoxidil: A Systematic Review and Individual Participant Data Meta-Analysis [PMID: 38885151 · 2025]
Kiratiwongwan R, Boonchai W, Kanokrungsee S · Dermatitis (2025)

Topical minoxidil is generally well tolerated, yet there have been a few reports of allergic contact dermatitis (ACD) confirmed through patch testing. This systematic review and individual participant data meta-analysis sought to elucidate the primary allergens in patients exhibiting ACD in response to topical minoxidil formulations and to ascertain the appropriate testing concentrations and vehicles of minoxidil itself. A comprehensive search was conducted across the PubMed, Scopus, and Embase databases utilizing the keywords "minoxidil" and "contact dermatitis," or "contact allergy," or "contact eczema." Studies documenting ACD in patients using topical minoxidil confirmed by patch testing were deemed eligible. Our analysis included 46 studies encompassing 99 patients with patch-test-confirmed ACD to minoxidil-based topical treatments. The majority of these patients (93.9%) were exposed to minoxidil without additional active components. Minoxidil itself was identified as the primary allergen in 74.7% of the patients, with propylene glycol being the next most common allergen at 17.1%. Other allergens identified included estradiol, butylene glycol, methylchloroisothiazolinone/methylisothiazolinone, canrenone, and latanoprost. The most effective concentration was found to be 2% minoxidil in propylene glycol, which yielded a 100% positivity rate. The findings indicate that minoxidil is the predominant allergen in ACD reactions to its topical formulations, followed by propylene glycol. For the accurate diagnosis of ACD related to minoxidil, patch testing with 2% minoxidil in propylene glycol is recommended, as are separate tests for propylene glycol and other potential allergenic ingredients.

Optimizing ultrasound-assisted extraction of bioactive compounds from Canthium horridum blume leaves utilizing polyols: A study on skin-related activities [PMID: 38774328 · 2024]
Myo H, Khat-Udomkiri N · Heliyon (2024)

The focus on reducing organic solvent usage, owing to their negative environmental and health impacts, is driving a search for innovative green alternative solvents in academia and industry. Canthium horridum Blume (CH) exhibits many therapeutic activities, including antioxidant and anti-inflammatory efficacy. The objective of this study is to evaluate the optimal solvent concentration using a simplex-lattice design with an aqueous-polyols mixture and to optimize the parameters for extracting bioactive compounds and antioxidant activities from ultrasound-assisted extraction (UAE) of CH leaves through central composite design (CCD) in response surface methodology (RSM). This study examines the total phenolic content (TPC), antioxidant activities, comparison of different extraction conditions, identification of bioactive compounds, cell cytotoxicity, cellular antioxidant activity, and melanin content reduction efficacy of the extracts. According to the findings from the simplex-lattice model, the ideal solvent composition consisted of 32.57%w/w butylene glycol, 32.92%w/w glycerine, and 34.51%w/w water. Furthermore, based on the response model, optimal extraction conditions were identified as a 15-min extraction time and a solvent-to-sample ratio of 32.94:1. In comparison to alternative extraction methods, ultrasonic-assisted extraction using the aqueous-glycerine-butylene glycol (GB-UAE) extract resulted in notably elevated TPC and antioxidant responses (p < 0.05). Major antioxidant bioactive compounds included 4-(Butoxymethyl) phenol, 3- O -Caffeoyl-4- O -methylquinic acid, Quercetin 3-(2G-glucosylrutinoside), 2,4-Dihydroxybenzoic acid and other bioactive compounds. The GB-UAE extract revealed greater cell viability than UAE using ethanol (EtOH-UAE) extract in both cytotoxicity and cellular antioxidant assays at the same concentration. Additionally, it exhibited comparable melanin content reduction efficacy at a higher concentration compared to that of EtOH-UAE extract. The researcher anticipates that the current study will advance the utilization of an aqueous-polyols system for extracting bioactive compounds extending beyond CH leaves. Although the potential applications of CH leaves in cosmetics and pharmaceutical formulations have been identified, further comprehensive mechanistic and clinical studies are required to fully understand their effects.

Metabolic engineering of Escherichia coli for enhanced production of 1,3-butanediol from glucose [PMID: 37783239 · 2023]
Islam T, Nguyen-Vo TP, Cho S, Lee J, Gaur VK, Park S · Bioresour Technol (2023)

1,3-Butanediol (1,3-BDO) finds versatile applications in the cosmetic, chemical, and food industries. This study focuses on the metabolic engineering of Escherichia coli K12 to achieve efficient production of 1,3-BDO from glucose via acetoacetyl-CoA, 3-hydroxybutyryl-CoA, and 3-hydroxybutyraldehyde. The accumulation of an intermediary metabolite (pyruvate) and a byproduct (3-hydroxybutyric acid) was reduced by disruption of the negative transcription factor (PdhR) for pyruvate dehydrogenase complex (PDHc) and employing an efficient alcohol dehydrogenase (YjgB), respectively. Additionally, to improve NADPH availability, carbon flux was redirected from the Embden-Meyerhof-Parnas (EMP) pathway to the Entner-Doudoroff (ED) pathway. One resulting strain achieved a record-high titer of 790 mM (∼71.1 g/L) with a yield of 0.65 mol/mol for optically pure (R)-1,3-BDO, with an enantiomeric excess (e.e.) value of 98.5 %. These findings are useful in the commercial production of 1,3-BDO and provide valuable insights into the development of an efficient cell factory for other acetyl-CoA derivatives.

Effects of 1,3-propanediol associated, or not, with butylene glycol and/or glycerol on skin hydration and skin barrier function [PMID: 37699769 · 2024]
Pinto JR, Monteiro E Silva SA, Leonardi GR · Int J Cosmet Sci (2024)

This study aimed to assess the effect of 1,3-propanediol at different concentrations (5%, 10%, or 15%), either applied alone or in combination with butylene glycol (BG) (5%) and/or glycerol (5%), on skin hydration and skin barrier function. The measurements were conducted using capacitance to determine skin hydration and trans epidermal water loss (TEWL) rates to evaluate skin barrier function. A total of 30 healthy female subjects participated in the study. Capacitance and TEWL measurements were conducted at multiple time points, including before application and at 15 min, 2 and 8 h after the humectants were applied to the forearms of the subjects. All the subjects provided written informed consent. The 1,3-propanediol in all concentrations and in all combinations (with BG and/or glycerol) increased skin hydration and improved skin barrier function 15 min, 2 and 8 h after application. Glycerol increased the hydration performance of 1,3-propanediol. The application of 1,3-propanediol at a concentration of 15%, either alone or in combination with other humectants, reduced the TEWL to a greater extent than lower concentrations of 1,3-propanediol. Furthermore, the addition of glycerol to 1,3-propanediol 15% improved the skin barrier and reduced TEWL when compared with 1,3-propanediol alone and with the combination of 1,3-propanediol + BG. The humectants significantly improved skin hydration and reduced TEWL throughout the 8-h time course. The increase in 1,3-propanediol concentration, as well as its combination with glycerol, provided a greater benefit to the skin, improving both hydration and the skin barrier function. Short-Chain Diol Effects on the Self-Assembly of Polyoxyethylene Oleyl Ethers in Dilute Aqueous Solutions and Their Cosmetic Applications [PMID: 42266070 · 2026]
Zhou J, Huang C, Meng T, Lai C · J Phys Chem B (2026)

In the present work, the effect of 1,2-propylene glycol (1,2-PG) or 1,3-butylene glycol (1,3-BG) incorporation on the self-assembling behaviors of polyoxyethylene oleyl ethers in dilute aqueous solutions is extensively investigated. This study aims to elucidate the mechanism by which the short-chain diol introduction contributes to preventing the phase separation occurring in aqueous makeup cleansing products under high temperatures. The critical micellization concentration and standard free energy of micellization of polyoxyethylene oleyl ethers in 1,2-PG-water mixed solvents steadily increase with increasing concentration of 1,2-PG in water, indicating that the addition to water of 1,2-PG disfavors micelle formation. Additionally, the introduction of 1,2-PG resulted in a significant increase in the effective surface area demand per molecule of polyoxyethylene oleyl ether, pointing to a reduction in the packing tightness of the surfactant molecules. Results of SAXS experiments indirectly demonstrate that the effective critical packing parameter of polyoxyethylene oleyl ethers in the aqueous solution of 1,2-PG decreases with increasing 1,2-PG concentration, which corresponds well with the changes in the cloud point of amphiphiles. NMR measurements confirm that these observations may be related to the swelling of the hydrophilic chain induced by excessive water penetration into the palisade layer, which helps counteract the dehydration of the POE (polyoxyethylene) segments induced by elevated temperatures. Furthermore, 1,3-BG exhibits effects similar to those of 1,2-PG but with a notably stronger intensity.

Formulation of Ascorbic Acid and Betaine-based Therapeutic Deep Eutectic System for Enhanced Transdermal Delivery of Ascorbic Acid [PMID: 38794349 · 2024]
Song JE, Jun SH, Ryoo JY, Kang NG · Pharmaceutics (2024)

L-ascorbic acid (AA), a potent antioxidant, is commonly used topically in the pharmaceutical and cosmetic fields. However, the incorporation of AA into topical formulations is difficult because of its highly unstable nature and relatively poor skin permeability. In this study, we propose an alternative strategy for improving the solubility and topical delivery of AA through its conversion to a therapeutic deep eutectic system (THEDES). AA and betaine (Bet)-based THEDESs were prepared at certain molar ratios and characterized using polarized optical microscopy, Fourier transform infrared spectroscopy, and differential scanning calorimetry. Solubility tests showed that AA in the form of THEDES was readily soluble in various polyols (glycerin, 1,3-butylene glycol, dipropylene glycol, and 1,3-propanediol) at a high concentration (approximately 40%). Furthermore, compared to AA alone or the physical mixture of AA and Bet, AA-based THEDES significantly enhanced AA delivery through porcine skin. In an in vivo human study, THEDES-containing serum reduced the markers of aging and induced an even skin tone. These findings indicate the utility of AA and Bet-based THEDES as novel transdermal delivery systems for AA. Furthermore, our approach also showed good extension to developing gluconolactone, a well-known natural antioxidant, and Bet-based THEDES, showing potential application in transdermal delivery systems.

Effect of Nitrosative Stress on Biofilm Formation and Upregulation of 2,3-Butanediol Dehydrogenase in Saccharomyces Cerevisiae [PMID: 41171461 · 2025]

Nitrosative stress is a phenomenon where reactive nitrogen species (RNS), oxidizes different cellular macromolecules. In this study we investigated the effect of sub-toxic dose of different nitrosative stress agents on S. cerevisiae grown with 2% ethanol as sole carbon source. Our SEM analysis showed significant increase in biofilm production under stress with changes in cellular morphology. Genes responsible for biofilm formation like FLO11, BSC2 and MAC1 in S. cerevisiae upon treatment with ac. NaNO 2 and SNP were also found to be upregulated. Redox enzymes like glutathione reductase (GR) showed an increase in specific activity in treated sets but catalase activity had no significant difference. Utilization of ethanol as sole carbon source was followed primarily by assessing the specific activity of Adh2p, which showed a significant 4-fold increase in both treated sets. This was also corroborated with gene expression analysis of ADH2. Together with this estimation of important enzymes of other associated metabolic pathways were also done to understand the changes in metabolic flux. Importantly both glyoxylate cycle and TCA cycle were found to be partially blocked under stress condition whereas, aldehyde reductase (Bdh2p) an important enzyme for 2,3-Butanediol production was found to have upregulated significantly. Altogether our study provides the first report on the effect of nitrosative stress on S. cerevisiae grown on ethanol as a carbon source with possibility to produce 2,3-Butanediol, an industrially important compound, that has a huge demand in the paint, drug and cosmetic industries.
