Preliminary evaluation of an extract of Amaranthus caudatus as a novel nutraceutical: effects on cholesterol and glucose absorption [PMID: 41601868 · 2025]
Mattiello A, Barbaglio G, Magnone M, Scarfì S, Vernazza S, Mirata S, Markus J, Letasiova S, Riva D, Premoli R · Front Nutr (2025)

The increasing prevalence of metabolic syndrome has prompted the development of nutraceuticals to support the management of glucose intolerance and dyslipidemia. Amaranthus caudatus is a potential source of bioactive compounds with metabolic relevance. A novel ingredient derived from Amaranthus caudatus seeds was characterized by untargeted metabolomic analysis and bioinformatic processing. Its effects were evaluated through in vitro enzyme inhibition and intestinal absorption assays, as well as preliminary observations in subjects with glucose intolerance over 3 months. The extract significantly inhibited key enzymes involved in carbohydrate metabolism and reduced glucose and cholesterol absorption in a three-dimensional intestinal model, while showing antioxidant activity. Preliminary human observations indicated improvements in HbA1c, post-prandial glucose, BMI, and a trend toward reduced total and LDL cholesterol. These results suggest that Amaranthus caudatus seed extract may represent a promising nutraceutical for supporting glycemic control and lipid metabolism.

Improved HDL, LDL and total cholesterol levels following a 3-month administration of Mentha spicata leaf extract and Amaranthus caudatus seed flour extracts, flavonoids and B vitamins. A placebo-controlled, double-blind, randomized clinical trial [PMID: 41519619 · 2026]
Di Minno A, Morone MV, Cordara M, Buccato DG, De Lellis LF, Ullah H, Piccinocchi R, Larsen DS, Baldi A, Piccinocchi G, Xiao X, Sacchi R, Daglia M · Nutr Metab Cardiovasc Dis (2026)

We have evaluated the efficacy and tolerability of two food supplements (FS) containing flavonoids (naringin and hesperidin); same doses of B3, B6, B9 and B12 vitamins, and two different doses of a blend of Mentha spicata leaf extract and Amaranthus caudatus seed flour, in subjects with borderline high total (TC) and low-density lipoprotein cholesterol (LDL-C) levels. 114 Participants (18-70 years) with TC levels 200-239 mg/dL, (5.18-6.19 mmol/L) and LDL-C (<159 mg/dL) were randomised into three groups to receive for 90 days the lowest (n = 38, Treatment A), the highest dose of the FS (n = 38 - Treatment B), or placebo (n = 38). Treatment B was associated with a significant reduction in LDL-C (∼31.5 mg/dL;-22%) and TC (∼19.5 mg/dL; -9%), along with an increase in high-density lipoprotein cholesterol (HDL-C). The greater efficacy of Treatment B containing the highest dose of vegetable extracts is likely attributable to its higher M. spicata extract content, as judged by high-resolution mass spectrometry analysis of the preparation. The combination of different FS ingredients with different mechanisms of action can be a valuable strategy for improving lipid profiles in subjects with borderline high TC and LDL-C levels.

Effect of Sprouting, Fermentation and Cooking on Antioxidant Content and Total Antioxidant Activity in Quinoa and Amaranth [PMID: 39456755 · 2024]
Vento M, Della Croce CM, Bellani L, Tassi EL, Echeverria MC, Giorgetti L · Int J Mol Sci (2024)

The study of different processing techniques, such as sprouting, cooking and fermentation, can help to develop new products for human health. In this work, raw, cooked and fermented seeds and germinated seeds of Chenopodium quinoa Willd. var. Tunkahuan and Amaranthus caudatus L. var. Alegrìa were compared for the content of antioxidant molecules, total antioxidant capacity and mineral elements. Fermentation was induced spontaneously, with the yeast Saccharomyces cerevisiae , with the bacterium Lactobacillus plantarum and with both microorganisms, for 24 and 48 h. The increase in antioxidant molecules and antioxidant activity was induced by germination, by 24 h of spontaneous fermentation (polyphenols and flavonoids) and by 24 h of L. plantarum fermentation (total antioxidant activity) for both species. Germinated seeds of the two plants showed higher values in respect to seeds of macroelements and microelements. No genotoxic but rather protective effects were determined for seed and germinated seed extracts using the D7 strain of S. cerevisiae , a good tool for the evaluation of protection from oxidative damage induced by radical oxygen species (ROS) in cells and tissues. Therefore, the two varieties could be very suitable for their use in human diet and in supplements, especially as germinated seeds or as fermented foods.

Obtaining biologically active substances from Amaranthus caudatus L. seeds in one technological cycle [PMID: 35466427 · 2022]
Korabel IM, Panchak LV, Zyn AR, Antonyuk VO · Biomed Chromatogr (2022)

Nowadays, amaranth is a valuable multipurpose crop and a source of a number of very important biologically active substances. The aim of this study was to develop a comprehensive scheme for obtaining fatty oil, triterpenoids and lectin from the seeds of Amaranthus caudatus L. in one technological cycle. Two variants of the lectin and triterpene compound purification method from amaranth seeds were tested. It was determined that the extraction of triterpene compounds should be carried out after purification of the lectin from degreased seeds. The rationality of this sequence of technological operations is explained by the lability of the lectin and the insolubility in water of triterpene compounds from amaranth seeds. The study also presents a scheme for obtaining squalene from amaranth oil by chromatography on silica gel and proposes a more effective affinity sorbent for purification of the lectin. The use of such a sorbent also opens up the possibility of preserving other water-soluble substances from amaranth seeds. The physicochemical characteristics and carbohydrate specificity of the lectin are described and new data on the results of the interaction of lectin with human and animal erythrocytes are given. The obtained results are discussed in the light of the complex use of raw materials.

Antioxidative activities and phenolic compounds of pumpkin (Cucurbita pepo) seeds and amaranth (Amaranthus caudatus) grain extracts [PMID: 28114838 · 2017]
Peiretti PG, Meineri G, Gai F, Longato E, Amarowicz R · Nat Prod Res (2017)

Phenolic compounds were extracted from pumpkin (Cucurbita pepo) seed and amaranth (Amaranthus caudatus) grain into 80% (v/v) methanol. The extracts obtained were characterised by the contents of total phenolic compounds (TPC), trolox equivalent antioxidant capacity (TEAC), ferric-reducing antioxidant power (FRAP) and antiradical activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH · ) radical. The content of individual phenolic compounds was determined by HPLC-DAD method. Pumpkin seeds showed the higher content of TPC than that from amaranth. The TEAC values of both extracts were similar each other. The lower value of FRAP was observed for pumpkin seed. Phenolic compound present in amaranth grain exhibited strongest antiradical properties against DPPH radical. Several peaks were present on the HPLC chromatograms of two extracts. The UV-DAD spectra confirmed the presence of vanillic acid derivatives in the amaranth grain. The three main phenolic compound present in pumpkin seed were characterised by UV-DAD spectra with maximum at 258, 266 and 278 nm.

Radical Scavenging Activities of Tannin Extracted from Amaranth (Amaranthus caudatus L.) [PMID: 25639718 · 2015]
Jo HJ, Chung KH, Yoon JA, Lee KJ, Song BC, An JH · J Microbiol Biotechnol (2015)

This study investigates the bioactivity of tannin from amaranth (Amaranthus caudatus L.) extracts. The antioxidant activities of the extracts from amaranth leaves, flowers, and seeds were evaluated. Tannin from leaves of amaranth has been evaluated for superoxide scavenging activity by using DPPH and ABTS(+) analysis, reducing power, protective effect against H2O2-induced oxidative damage in L-132 and BNL-CL2 cells, and inhibition of superoxide radical effects on HL-60 cells. At a concentration of 100 μg/ml, tannin showed protective effects and restored cell survival to 69.2% and 41.8% for L-132 and BNL-CL2 cells, respectively. Furthermore, at the same concentration, tannin inhibited 41% of the activity of the superoxide radical on HL-60 cells and 43.4% of the increase in nitric oxide levels in RAW 264.7 cells. The expression levels of the antioxidant-associated protein SOD-1 were significantly increased in a concentration-dependent manner in RAW 264.7 cells treated with tannin from amaranth leaves. These results suggest that tannin from the leaves of Amaranthus caudatus L. is a promising source of antioxidant component that can be used as a food preservative or nutraceutical.

In Vitro antioxidant effect and inhibition of alpha-amylase of two varieties of Amaranthus caudatus seeds [PMID: 15930754 · 2005]
Conforti F, Statti G, Loizzo MR, Sacchetti G, Poli F, Menichini F · Biol Pharm Bull (2005)

This study reports on the biological properties, antioxidant and antidiabetic, of two varieties of Amaranthus caudatus seeds, Oscar blanco and Victor red. Oil, squalene and phenolic contents were also determined. Seeds of both investigated varieties were found to possess very different levels of squalene (2.2% in Oscar blanco variety and 7.5% in Victor red variety). Although the antioxidant activity of A. caudatus var. Oscar blanco and A. caudatus var. Victor red statistically did not differ significantly from each other (IC50 values of ethyl acetate extracts were 0.50 mg/ml and 0.62 mg/ml, respectively), significant differences were noticed in relation to antidiabetic activity (inhibition of alpha-amylase, EC 3.2.1.1) of methanolic extracts that showed 50.5% for A. caudatus var. Oscar blanco and 28% for A. caudatus var. Victor red at concentration of 25 microg/ml.

Rapid techniques for the extraction of vitamin E isomers from Amaranthus caudatus seeds: ultrasonic and supercritical fluid extraction [PMID: 12918869 · 2002]
Bruni R, Guerrini A, Scalia S, Romagnoli C, Sacchetti G · Phytochem Anal (2002)

Supercritical fluid extraction (SFE) of seeds of Amaranthus caudatus (Amaranthaceae) and the use of ultrasound as a co-adjuvant in the extraction process were compared with methods traditionally used in the extraction of tocopherols and fatty acids. The use of readily available ultrasound equipment as an adjunct to the classical methods employed for the extraction of tocols provided qualitatively acceptable results more rapidly and more economically. SFE gave quantitatively better yields in shorter times, with solvent-free extracts obtained under conditions that minimised the degradation of thermolabile components. No significant variations were observed in the profile of the fatty acids extracted from amaranth oil by SFE or other methods, thus confirming the qualitative comparability of the faster supercritical extraction with the more time-consuming classical techniques even when processed with the aid of ultrasound.

Antioxidant activity of ethanolic extracts of amaranth seeds [PMID: 12108218 · 2002]
Klimczak I, Małecka M, Pachołek B · Nahrung (2002)

Antioxidant activity of ethanolic extracts obtained from two amaranth species was evaluated in a beta-carotene-linoleic acid model system. The addition of amaranth extracts in the range of 0.01-0.1% inhibited degradation of a beta-carotene in a model emulsion during incubation at 60 degrees C; 0.05% addition of amaranth seeds extract was proposed as practically applicable. The total content of phenolic compounds was estimated by the Folin-Ciocalteu method and ranged from 39.17 mg/100 g of Amaranthus caudatus to 56.22 mg/100 g of A. paniculatus seeds. Free phenolic acids contained in ethanolic extracts of amaranth seeds were purified and isolated by solid-phase extraction (SPE) and identified by reversed-phase high-performance liquid chromatography (RP-HPLC). The technique involved gave a good separation of the free phenolic acids in the amaranth seeds. Significant differences in phenolic acids profiles of both amaranth species were observed.

Wild Amaranthus caudatus seed oil, a nutraceutical resource from Ecuadorian flora [PMID: 11714343 · 2001]
Bruni R, Medici A, Guerrini A, Scalia S, Poli F, Muzzoli M, Sacchetti G · J Agric Food Chem (2001)

Seed oil of wild Amaranthus caudatus from Ecuador was analyzed for determining the tocopherol, fatty acid, and sterol contents. The data obtained were compared with the analogous chemical profile of seed oil of Italian A. caudatus with the objective of evaluating the nutraceutical and alimentary potential of the Ecuadorian matrix. Supercritical fluid and ultrasound-enhanced extractions were performed on both matrices. Qualitative and quantitative determinations of tocopherols were performed by HPLC, whereas GC and GC-MS were used to determine the fatty acid composition and sterols, respectively. Supercritical fluid extraction at 400 atm was the most efficient extraction method in terms of both total yield extract and tocopherol yield. Seeds of Ecuadorian of A. caudatus contained higher levels of tocopherols than Italian samples, whereas the fatty acid composition and sterol content were similar. From the obtained results it can be suggested that seed oil of wild Ecuadorian A. caudatus can prove to be an effective nutraceutical and alimentary resource and a valid alternative to the European varieties.
